abstract | Highly productive recombinant yeast strains with modified galactose-regulated transcription, and use thereof . The object of this invention are highly-productive recombinant organisms with galactose-regulation, in which on chromosomes the gene encoding the enzyme for the metabolic conversion of the transcription inductor into an inactive product is replaced with a gene encoding the transcription factor for the galactose-inducible promoter. The isogenic strain is preferably chosen from Saccharomycetaceae or Cryptococcaceae families, more preferably the Saccharomyces genus. The GAL1 gene encodes the enzyme for the galactose metabolism, and the GAL4 gene encodes the transcription factor. The galactose-inducible promoter is regulated by the Gal4 transcription factor, and regulates the transcription of GAL1, GAL2, GAL7, GAL10 or MEL1. The claimed strains are hosts for plasmids applicable in heterologous gene expression from the galactose-inducible promoter, and are used in the production of bioactive substances, and other products of biotechnology. The object of this invention is the production of bioactive substances, and other products of biotechnology, using above described strains. |
