abstract The invention relates to a method of determining the mutational load of a gene library obtained by random mutagenesis of a particular gene of interest and means for implementing same, comprising the following steps: a) preparation of an abacus that links the mutational load (CM) of a gene library obtained by random mutagenesis of a model gene to the fraction of mutated model genes observed in said library; b) random mutagenesis of the model gene that was used to prepare the abacus in step (a) and of the gene of interest, in order to produce the corresponding mutated gene libraries; c) determination of the mutational load (CM) of the gene library obtained from the model gene in step (b) using the abacus established in step (a); and d) application of a correction factor (CF) to the mutational load (CM) of the mutated model gene library determined in step (C), in order to determine the mutational load (CM) of the library of mutated genes of interest from step (b).
