abstract The present invention relates to a method for activating a nucleic acid for a strand displacement reaction comprising the steps of: (a) heating a nucleic acid (preferably a double-stranded DNA) to a temperature of 50 to 90 ° C, (b) cooling the nucleic acid a temperature at which a polymerase does not show a significant decrease in activity, and (c) starting the strand displacement reaction by adding a heat labile polymerase to the nucleic acid. In an alternative method employing a heat stable polymerase, the polymerase may be added immediately in step (a). The method can be carried out both with purified nucleic acid and nucleic acid contained in a cell.
