abstract It is intended to specify a region of a wheat endogenous DNA (genome) with which wheat can be specifically detected in a single copy without any cross-over with other plants in PCR and provide primers for amplifying this region and a method of appropriately detecting and quantifying the endogenous DNA by using the same. Namely, a method of detecting or quantifying a wheat endogenous DNA sequence in a test sample by the polymerase chain reaction which involves the step of using a nucleic acid in the test sample or a nucleic acid extracted from the test sample as a template and amplifying a nucleic acid in a region containing at least 80% of a base sequence represented by any of SEQ ID NOS:1 to 7 with the use of a pair of primers capable of amplifying this region; and the step of detecting or quantifying the thus amplified nucleic acid.
