abstract | The present invention relates to a method for producing a functional human immunoglobulin, wherein a human heavy chain immunoglobulin, devoid of any light chain, is expressed, comprising the steps of: a) transforming a filamentous host cell with a recombinant construct encoding a modified human heavy chain immunoglobulin, wherein the modifications comprise one or more mutations in the region of the heavy chain protein involved in contact with the light chain; b) culturing said filamentous host cell under conditions promoting expression of said modified human heavy chain immunoglobulin; and c) recovering said modified human heavy chain immunoglobulin. |
