abstract The present invention is aimed at providing the aminonacid sequence of a nitrile hydratase having a novel mutationnpoint, and the base sequence of the gene. It is also aimednat providing a method for modifying an enzyme having annitrile hydratase activity. n As a means to achieve the above objects, the amino acidnsequence of a mutant which is obtained by introducing annovel mutation into a Pseudonocardia thermophila JCM3095-derivednnitrile hydratase consisting of two types ofnheterogeneous subunits, and the base sequence of the genenare provided. Further, the nitrile hydratase is modified bynspecifying the region to be modified in thenstereostructure/amino acid sequence of the nitrile hydratase,nand applying alteration such as substitution, insertion,ndeletion or the like, to the amino acids in the amino acidnsequence which are corresponding to the amino acid residuesnforming the region. n According to the invention, it is possible to convert annitrile compound to a corresponding amide compound morenefficiently compared with the prior art.
