abstract | The invention provides an emulsion-based method for thenpreparation of controlled release microspheres for the delivery of activencompounds. The method comprises the preparation of an emulsion comprisingnan aqueous dispersed phase which comprises a polymer capable of forming anhydrogel, a bioactive protein, and water, and which is substantially free fromninsoluble aggregates of the bioactive protein. Subsequently, the polymernphysically or chemically crosslinked to form a hydrogel. The invention furthernprovides active protein-loaded hydrogel microspheres which are prepared bynthe process, and which are substantially free from insoluble aggregates of thenactive protein. The microspheres exhibit controlled release, with releasenprofiles which are considerably improved over those of previously knownnhydrogel microspheres. The microspheres may be used to deliver therapeuticnor diagnostic proteins by injection. |
