abstract | The invention concerns a process for producing transgenicneucaryotic cells, particularly plants, which comprises:ncontacting a culture of untransformed cells with an inhibitornof poly-(ADP ribose) polymerase for a period of timensufficient to reduce the response of the cultured cells tonstress and to reduce their metabolism. The untransformedncells are then contacted with foreign DNA comprising at leastnone gene of interest under condtitions in which the foreignnDNA is taken up by the untransformed cells and thhe gene ofninterest is stably integrated in the nuclear genome of thenuntransformed cells to produce the transgenic cells.nOptionally, the transgenic cells are recovered from thenculture. Preferably, the inhibitor is niacinamide, preferablynat a concentration of about 200mg/l to 500 mg/l and thenuntransformed cells are cultured in a medium containing theninhibitor for a period of time of approximately 3 to 14 daysnprior to the contacting with the foreign DNA. The inventionnalso relates to a plant having in the nuclear genome of itsncells foreign DNA integrated only in the regions of thennuclear genome that are transcriptionally active in cells ofnthe plant wwhen the cells are treated with an effectivenamount of a PARP inhibitor for a period of time ssufficientnto reduce cell metabolism to a state where gene expression isnessentially limited to genes expressed irrespective of thendifferentiated or physiological condition of the cell. |
