abstract | A molecule assigning a genotype to a phenotype, whichncomprises a nucleic acid portion having a nucleotidensequence reflecting the genotype, and a protein portionncomprising a protein involved in exhibition of the phenotype,na 3'-terminal end of the nucleic acid portion and a C-terminalnend of the protein portion being covalently bound,nand a method for constructing the molecule for assigningnthe genotype to the phenotype, which comprises (a) preparingna DNA containing a gene which has no termination codon, (b)ntranscribing the prepared DNA into RNA, (c) bonding anchimeric spacer composed of DNA and RNA to a 3'-terminalnend of the obtained RNA, (d) bonding, to a 3'-terminal endnof the obtained bonded product, a nucleoside or a substancenhaving a chemical structure analogous to that of a nucleoside,nwhich can be covalently bound to an amino acid or a substancenhaving a chemical structure analogous to that of an aminonacid, and (e) performing protein synthesis in a cell-freenprotein synthesis system using the obtained bonded productnas mRNA to bond a nucleic acid portion containing the genento a translation product of the gene. The moleculenassigning the genotype to the phenotype of the presentninvention is an extremely useful substance that can be usednfor evolutionary molecular engineering, i.e., modificationnof functional biopolymers such as enzymes, antibodies, andnribozymes, and creation of biopolymers having functionsnwhich cannot be found in living organisms. |
