abstract |
The present invention relates to engineeringnattenuated viruses by altering a non-coding region ornthe coding sequence of a viral gene. Alterations ofnthe non-coding regions which regulate transcriptionnand/or replication are described. These alterationsnresult in the down-regulation of the viral gene andnan attenuation of the virus, either by the productionnof defective particles during replication, or bynreducing the number of progeny virions producednduring viral replication. Alterations of viral codingnsequences are also described which result in anrecombinant or chimeric attenuated virus. |
