abstract | Antibodies against RNA polymerase (RNAP) are frequently detected in patients suffering from diffuse scleroderma showing the rapid progress of sclerema and relate to a disease type accompanied by sclerodermatous renal crisis as a complication and showing extremely poor prognosis. Thus, it is intended to provide a method of conveniently detecting anti-RNAP antibodies which are highly useful in diagnosing scleroderma, classifying disease types and presuming organ disorders, in particular, sclerodermatous renal crisis. To identify an epitope recognized commonly by anti-RNAP antibodies, the full-length RPC62 and a fragment of RPC155, which are 2 subunits of RNAPIII of 62 kDa and 155 kDa respectively, are expressed as a recombinant protein in Escherichia coli. Then the reactivities with anti-RNAP antibody-positive and negative patients suffering from scleroderma are examined by the immunoblotting method so as to confirm that an epitope which is recognized commonly by anti-RNAP antibodies in the sera of the patients suffering from scleroderma exists in the amino acid residues at the 891- to 1020-positions of the RPC155. |
