abstract | The invention relates to a method for determining the binding capability of a structuren(ligand) to an analyte, which method comprises:n i. applying the sample to an application zone for sample (ASZ) on a flownmatrix in which transport of components present in the sample may takenplace (transport flow), the flow matrix further comprising:n a) optionally an application zone (AR*Z) for a binding reactantn(Reactant* = R*) which is analytically detectable, b) a detection zone (DZ) which is located downstream of ASZ andncomprises another binding reactant (Capturer) firmly anchored to thenmatrix and in which a complex (signal complex) containing thenCapturer and the analyte and/or Reactant* is formed during thenmethod, and ii. detecting the signal complex in the detection zone, the measured signalnbeing used for determining the analyte, nwherein the flow matrix comprises at least one separation zone (SZ) between ASZ andnDZ and this zone (SZ) exhibits a structure (ligand) having binding capability for thenanalyte, and the binding capability of the ligand for the analyte is determined. Accordingnto the invention, the flow matrix comprises at least one separation zone (SZ) betweennASZ and DZ, which zone exhibits a structure (ligand) having binding capability for thenanalyte that is transported in the matrix. The invention also relates to a test kitncomprising the flow matrix. |
