abstract The invention relates to <i>Saccharomyces-cerevisiae</i> yeast strains with a stable integration and expression of the nucleic acid sequence for a heterologous potassium ion channel, in addition to a method for constructing said yeast strains. In particular, the invention relates to a <i>Saccharomyces-cerevisiae</i> yeast strain, in which all identified potassium ion translocation systems have been specifically deleted and to a <i>Saccharomyces-cerevisiae</i> yeast strain, into which in addition a nucleic acid sequence, which codes for a heterologous potassium ion channel, in particular for a mammalian potassium ion channel such as the human erg potassium ion channel (HERG), is integrated in a stable manner into the yeast genome and is expressed. The invention also relates to a method for the stable integration of heterologous potassium ion channel proteins into a <i>Saccharomyces-cerevisiae</i> yeast strain and to a method for detecting specific modulators of mammalian potassium ion channels, using said yeast strains.
