abstract | The present invention relates to a method and a kit for isolating nucleic acids from tissue samples. In particular, but not exclusively, formalin fixed paraffin embedded tissue samples. The isolated nucleic acids will normally be for use in a polymerase chain reaction (PCR). The inventive method comprises the sequential steps of: a) placing in a container the tissue sample, and a wax, a detergent and a tissue digestion enzyme; b) heating the container to a first temperature; c) raising the temperature of the container to a second temperature; d) cooling the container; e) forming a waxy-layer containing impurities and a nucleic acid containing layer; f) removing the waxy-layer, such that the nucleic acids are usable in a polymerase chain reaction. Kits are also provided for performing the same. |
