abstract | A genetically modified thrombin which is efficiently produced with the use of a gene modification technique which comprises: 1) the step wherein a transformant constructed by transforming an animal cell with an expression vector carrying a gene encoding prethrombin integrated into the downstream of a promoter is cultured and the prethrombin thus produced and accumulated in the culture medium is collected; 2) the step wherein the human prethrombin-containing solution thus collected and recovered is activated by treating with ecarin to thereby convert into human thrombin; and 3) the step wherein purified human thrombin is obtained from the liquid reaction mixture having been inactivated as described above. Thus, human thrombin can be provided in a large amount at a high safety and a low cost while minimizing the effects of blood-origin components. |
