abstract Methods, vectors, cell lines and kits for the screening and identification of nucleic acids are described. The invention is based on the use of a site-specific recombinase to excise, from an expression vector into which an exogenous nucleic acid having a desired feature has been inserted, a region of the vector which is excisable by site-specific recombination. Insertion into the vector of a nucleic acid having a desired feature, such as nucleic acids capable of changing the expression of cellular genes or the state of cellular metabolism or signaling pathways, triggers the synthesis and/or activity of a site specific recombinase, the action of the recombinase allowing an easy selection of the expression vector containing the exogenous nucleic acid having a desired feature.
