abstract Disclosed is a method for determining whether a first proteins capable of physicallyninteracting with a second protein. The method involves: (a) providing a hostncell which contains (i) a reporter gene operably linked to a protein binding site;n(ii) a first fusion gene which expresses a first fusion protein, the first fusion proteinnincluding the first protein covalently bonded to a binding moiety which isncapable of specifically binding to the protein binding site; and (iii) a second fusionngene which expresses a second fusions protein, the second fusion protein includingnthe second protein covalently bonded t a weak gene activating moiety; and (b)nmeasuring expression of the reporter gene as a measure of an interaction betweennthe first and the second proteins. Such a determination facilitated the isolation ofnthe gene encoding the interacting protein. Also disclosed herein is recombinantnCdil polypeptide, nucleic acid encoding the Cdil polypeptide, and uses thereof.
