abstract | The invention relates to a method for increasing the copy number of a chromosomally integrated expression cassette in a microbial strain without leaving antibiotic resistance markers behind in the strain, the necessary genetic constructs, and the strains resulting from the method of the invention. In the method an expression cassette comprising a gene of interest and a copy of a gene being non-functional in the chromosome of the host cell is introduced in the host cell. The host cell is cultivated in the presence of a precursor to an inhibiting compound produced if the expression cassette is not integrated into the chromosome. |
