abstract | According to the present invention, a method wherein anpancreas induced in vitro which enables to obtain knowledgenregarding the mechanisms of differentiation and formation ofnpancreas that is useful for developmental engineering and organnengineering, a pancreas for transplantation by which it can benevaluated whether or not a pancreas induced in vitro cannfunction in practice in vivo,and a pancreas induced in vitronwhich contributes to the development of diagnosis and treatmentnof pancreas disorder for higher animals, can artificially andnefficiently be induced from a gastrula apart from thenpresumptive region of pancreas is provided. Presumptivenectoderm region of undifferentiated cells of late blastula thatnforms atypical epithelium but not pancreas when cultured innvitro, was cut out from a late blastula of a Xenopus, treatednwith acitivin, followed by treatment with retinoic acid afternthree to five hours. These explants were subjected tonstationary culture in Steinberg's solution containing BSA, andnits developmental fate was changed to pancreas, forming anmorphologic as well as functional pancreas in vitro with highnefficiency. |
