abstract |
This method permits minority genomes to be detected, more specifically, minority memory genomes of a nucleic acid population present in less than 50% of a viral quasispecies and that contain at least one mutation in comparison to the majority genome of the quasispecies and consists in: extraction of the nucleic acid from a sample suspected to contain this viral quasispecies; b) amplification of at least one nucleic acid fragment from this quasispecies; and c) detection and analysis of the existence of minority genomes using techniques based on the use of DNA microchips, Heteroduplex trace assay (HTA) and molecular cloning. The method can be used in genetic diagnostics of viral diseases. |
