abstract | The invention relates to a method for the production of one or several proteins in a host cell of the yeast genus Arxula and a nucleic acid molecule, comprising a suitable promoter. The invention further relates to an expression vector, a host cell, a kit and the use thereof. The aim of the invention is a method for the production of a protein in a yeast of the genus Arxula, by means of which large amounts of protein may be obtained. Said aim is achieved by means of a method for the production of one or several proteins in a host cell of the yeast genus Arxula, comprising: (i) cloning of at least one nucleic acid, which codes for a heterologous protein in an expression vector, containing an inducible promoter from a yeast of the genus Arxula, or a promoter from a constitutively expressed gene, chosen from the ARFC3 gene and the AHSB4 gene, from a yeast of the genus Arxula, such that the cloned nucleic acid is under the transcription control of the promoter; (ii) introduction of the expression vector obtained in (i) into a host cell of the yeast genus Arxula, suitable for the production of the heterologous protein; (iii) cultivation of the host cells obtained in (ii) and, optionally, (iv) induction of the promoter; (v) harvesting the protein by a known method. |
