abstract | A method for preparing a cDNA from a mRNA using a reversentranscriptase wherein reverse transcription is performed at antemperature at which the mRNA does not take a secondary structure,nfor example, at a temperature of 45°C or more. The method isnperformed, for example, using a heat-labile reversentranscriptase in the presence of a substance exhibitingnchaperone function having chaperone function such asnsaccharides. The method is performed, for example, in thenpresence of metal ions necessary for activation of the reversentranscriptase and a chelating agent for the metal ions such asna deoxynucleotide triphosphate. The method is capable ofnreverse transcription over the full length of mRNA template evennif the mRNA is a long chain mRNA and, as a result, producingna full length cDNA. |
