Provided are polynucleotides encoding Pyruvate:NADP+ oxidoreductasesn(PNO) as well as methods for obtaining the same. Furthermore,nvectors comprising said polynucleotides are described, whereinnthe polynucleotides are operatively linked to expressionncontrol sequences allowing the expression in prokaryotic and/orneukaryotic host cells. In addition, polypeptides encoded by saidnpolynucleotides, antibodies to said polypeptides and methods forntheir production are provided. Further described are methods fornincreasing the acetyl CoA synthesis as well as methods for thenproduction of fatty acids, carotenoids, isoprenoids, vitamins,nlipids, wax esters, (poly)saccharides and/or polyhydroxyalkanoates,nor its metabolism products, in particular, steroid hormones,nprostaglandin, cholesterol, triacylglycerols, bile acids ornketone bodies, comprising the expression of the polynucleotide ornpolypeptide described herein in a host cell or plant cell, plantntissue or plant. Methods for the identification of compoundsnbeing capable of activating or inhibiting PNO are described asnwell. Further, a pharmaceutical composition comprising the aforementionedninhibiting compounds and antibodies is described.
