titleOfInvention | ENZYMATIC FLUORIMETRIC ASSAY OF cAMP AND ADENYLATE CYCLASE |
abstract | The present invention relates to a method for quicklyndetermining cAMP content or an adenylate cyclase activitynin a biological sample containing non-cyclic adeninennucleotides without the use of radioactive agents. n Particularly, the present invention provides a methodnof determining cAMP content or an adenylate cyclasenactivity in a biological sample containing non-cyclicnadenine nucleotides selected from the group consisting ofncAMP produced by endogenous adenylate cyclase, and AMP, ATP,nADP and a mixture thereof, which comprises (1) combining anbiological sample with effective amounts of apyrase, nadenosine deaminase and alkaline phosphatase tonenzymatically remove non-cyclic adenine nucleotides othernthan cAMP, and glucose-6-phosphate in the sample; (2)nenzymatically converting cAMP into AMP; (3) determining annamount of AMP without the use of radioactive agents, and ankit to carry out the method. |
