abstract |
Retroviral vectors are disclosed which are useful in gene therapy. The packageablenRNA genome of the vector provides in the DNA provirus a regulated promoternwhich is inducible by a regulatory factor e.g the HIV transactivator protein Tat,nand at least one selected gene under its transcriptional control. In the DNAnprovirus, the regulated promoter is present in the 5' long terminal repeat (LTR) innplace of the 5' LTR promoter function of the retrovirus, and the selected gene isnlocated between the LTRs. Thus when inserted into a host cell the gene will benexpressed only when the DNA provirus is exposed to the regulatory factor. |
