abstract | Methods are disclosed for analysing a sample to determine the presence or otherwise therein of a target polynucleotide sequence. The method involves treating the nucleic acid under hybridising conditions with first and second polynucleotide probes adapted to bind to adjacent mutually exclusive regions of the target sequence (if present). Each of the probes carries a different chromophoric moiety which is capable, with the other moiety, of forming an exciplex on photo-irradiation if the two probes are hybridised to the target sequence. Methods are disclosed for enhancing the exciplex emission and include the possibilities of (i) the first and second moieties coming into exciplex forming relationship in a localised region of greater hydrophobicity than the bulk phase of the sample being analysed; (ii) the first and second moieties coming into exciplex forming relationship within the immediate vicinity of the double stranded nucleic acid; (iii) there being included in the sample being analysed a compound capable of developing an exciplex fluorescent signal when the first and second moieties are in exciplex forming relationship; (iv) applying a magnetic field to enhance exciplex emission; and (v) the use of at least one of the steps, in either order, of (a) at least partial removal of relatively polar medium, and (b) addition of a less polar medium. |
