Transforming growth factor β (TGF-β) isnproduced in relatively large quantities and at anrelatively high purity by fermentation in a perfusionnmicrocarrier reactor. Conditioned media from the reactornis first treated to provide the active form of TGF-β andnsubsequently purified by cation exchange chromatographynfollowed by hydrophobic interaction chromatography.nOptionally, nucleic acids complexed with the TGF-β may benremoved while the protein is bound to the cation exchangenresin. Purified TGF-βI compositions have a specificnactivity above 10 7 U/ml. |
