abstract Human minor histocompatibility Ags (mHag) play an important'nrole in the induction of CTL reactivity against tumours afternHLA-identical allogeneic bone marrow transplantation (BMT).nAs most mHag are not tumour specific but also expressed bynnormal tissues, anti-tumour reactivity is often associatednwith life-threatening graft-versus-host disease (GVHD). Here,nwe describe the use of newly found tumour specific mHag fornthe development of novel means and methods for the treamentnof cancer. n In one example we identified a novel mHag, HB-1, that elicitsndonor-derived CTL reactivity in a B-cell acute lymphoblasticnleukemia (B-ALL) patient treated by HLA-matched BMT. Wenidentified the gene encoding the antigenic peptide recognisednby a HB-1 specific CTL. Expression of the HB-1 gene was onlynobserved in B-ALL cells and EBV-transformed B cells. Analysisnrevealed that the HB-1 gene is polymorphic. The restrictednexpression of the polymorphic HB-1 Ag by B-ALL cells was usednin one example of the invention to generate HB-1 specific CTLnin vitro using peptide-loaded dendritic cells. HB-1 specificnCTL against B-ALL were used to combat B-ALL and were shown tonat least in part reduce the growth of said leukemia withoutnevoking GVHD.
