abstract The present invention relates to develop a series of B- and T-cell clonality assay kits. The data presented here has shown the importance of the B- and T-cell clonality assay kits in the early diagnosis and differential diagnosis for MS, PCNSL and other neurological diseases. In comparison to current diagnostic technologies (cytology, oligoclonal bands, CT, MRI, and surgical biopsy), the invented technology demonstrates its benefit as a technique which is high sensitive, specific, non-invasive, and economical, and which can be applied to different specimens (CSF cells, surgical biopsy and autopsy). Utilization of this technology to MS study has shown that the B-cell clonal expansion is present in majority of MS patients, which have undergone antigen-driven somatic mutation in the germinal centers of peripheral lymphoid tissues. This finding implicates an important pathogenic pathway for the development of demyelination in CNS of MS. Furthermore, this technology allows the establishment of clonal specific RNA library from pathogenic cells in the CNS of the patients, which is important for further understanding of the role of antigen(s) in the cause of B-cell clonal expansion, and towards to developing antigen specific therapeutic strategy.
