abstract Disclosed is a method for producing double-stranded DNAncomprising treating double-stranded DNA having a homopolymernpart or parts at one or both ends with a restriction enzymento partly or fully eliminate at least one of the homopolymernpart or parts. The restriction enzyme is capable of cleavingndouble-stranded DNA at a cleavage site separate from anrecognition site therefor. Disclosed is a method forndetermining a nucleotide sequence of double-stranded DNAnutilizing one or both strands of the double-stranded DNA asna template, wherein the double-stranded DNA used as thentemplate is double-stranded DNA prepared by the above methodnof the present invention. The present invention provides anmethod for producing double-stranded DNA a part or all of whichnhomopolymer part, which may inhibit nucleotide sequencendetermination, is eliminated, and a method for determining annucleotide sequence utilizing as a template the double-strandednDNA prepared by the above method of the presentninvention, whose homopolymer part is partially or fullyneliminated.
