abstract | Antiinflammatory agents which are excellent in pharmacological effects and exhibit selective COX-2 inhibiting activity leading to an expectation of reduction in the adverse effects such as gastric mucosal disturbances. Compounds of general formula (I) have excellent antiinflammatory activity, and exhibit potent inhibitory action against COX-2 induced at an inflamed site with little inhibitory action against COX-1 always present in the stomach, kidney and so on, (wherein R<1> is hydrogen or halogeno; R<2> and R<3> are each hydrogen, halogeno, lower alkyl, lower alkoxy, hydroxyl or lower alkanoyloxy; R<4> is halogenated lower alkyl or lower alkyl; X is sulfur, oxygen or NH; and Y is lower alkylthio, lower alkylsulfonyl or sulfamoyl). Accordingly, drugs containing the compounds are useful as antiinflammatory agents reduced in the adverse effects such as gastric mucosal disturbances by virtue of the selective inhibitory activity of the compounds against COX-2. <CHEM> |
