abstract |
Vesicle (A, designated texosome) derived from tumour cells (a) is separated from its natural environment and comprises a lipid bilayer surrounding a cytosolic fraction (CF) or (b) has diameter 60-90 nm and presents at its surface: (i) tumour-specific antigenic peptides associated with MHC (major histocompatibility complex) Class I and/or II molecules; and (ii) molecules (I) for co-stimulation of lymphocytes. Also new are: (1) antigen-presenting cells (APC) loaded with (A); (2) membrane vesicles (B) separated from their natural environment, secreted by the APC of (1); (3) membrane vesicles (C, designated dexosomes) derived from a dendritic cell (DC) and containing at least one each of MHC Class I and Class II molecules; (4) combination of texosomes and/or dexosomes with an immunostimulating adjuvant for use simultaneously, separately or staged in time; and (5) use of interleukin (IL)-10 and/or IL-12 for production of dexosomes. |
