abstract The present invention relates to an induciblenpromoter gene for use in the fission yeastn Schizosaccharomyces pombe , an inducible expression vectorncontaining the inducible promoter gene,n Schizosaccharomyces pombe transformed with the induciblenexpression vector and a process for producing anheterologous protein by using the Schizosaccharomycesnpombe transformant. n The present invention also relates to a secretionnsignal gene for use in Schizosaccharomyces pombe , annexpression vector containing the secretion signal,n Schizosaccharomyces pombe transformed with the expressionnvector and a process for producing a heterologous proteinnby using the Schizosaccharomyces pombe transformant.nThese inducible promoter gene and secretion signal genenare originated from the S. pombe gene involved innexpression of the invertase precursor. n The inducible promoter gene is originated from thenregion responsible for the catabolite repression of theninvertase precursor. When a Schizosaccharomyces pombe ntransformant carrying the inducible expression vector isnincubated, the timing of the production of the desirednheterologous protein can be controlled by regulating thenexpression of the heterologous protein structural genendepending on the presence or absence of a specificnnutrient (especially glucose). n The secretion signal gene encodes the secretionnsignal for the invertase precursor. When an Schizosaccharomyces pombe transformant carrying annexpression vector containing the secretion signal gene isnincubated, a heterologous protein fused with thensecretion signal is synthesized as a precursory proteinnin the cell, and after intracellular separation of thensecretion signal, the heterologous protein is secretednfrom the cell. Therefore, it is possible to obtain thendesired protein from the cell culture, and the desirednheterologous protein can be purified easily.
