abstract |
A monoclonal antibody specific to Nap was prepared by usingncrude membrane fraction from cell line which has highnaffinity to Nef as an antigen. A cDNA clone which encodesnNap was obtained by screening a cDNA library of said cellnline utilizing the specific antibody. The nucleotidensequence of this cDNA was elucidated whereupon the full aminonacid sequences of human Nap was identified. n Since the anti-Nap monoclonal antibody inhibits the bindingnof Nef to Nap, it can be used as a new therapeutic agent fornAIDS. In addition, it can be applied for the diagnosis ofnAIDS by clarifying the relation between the expression of Napnand the development of AIDS. Nap can be utilized for bindingnexperiments to Nef. It is useful for the development ofnnovel therapeutic agents for AIDS based upon a mechanism ofnaction which inhibits the attachment of both factors andndevelopment of new inhibitors against the onset of AIDS by anmechanism of action different from that of reversentranscriptase inhibitors or protease inhibitors. |
