abstract | a) Fumarsäure in einem inerten Verdünnungsmittel in Gegenwart von Aspartasenoder Aspartase-produzierenden Mikroorganismen mit einem Überschuß annAmmoniak zu Ammonium-L-Aspartat umgesetzt, anschließend b) die Ammonium-L-Aspartat hältige Lösung mit Natriumhydroxid versetzt wird,nwobei sich je nach eingesetzter Menge an Natriumhydroxid das Mono- odernDinatrium-L-Aspartat oder ein Gemisch davon bildet und c) der sich abspaltende und überschüssiger Ammoniak bei Normaldruck undneiner Rückflußtemperatur von 90 - 120°C unter Verweilzeiten von 0,2 bis 2nMinuten kontinuierlich destillativ entfernt und in eine Fumarsäuresuspension,ndie als Ausgangslösung für weitere enzymatische Umsetzungen verwendetnwird, rückgeführt wird, worauf d) die verbleibende Natrium-L-Aspartat hältige Lösung mit Benzyloxycarbonylchloridnbei einem pH-Wert zwischen 9 und 14 bei gleichzeitiger Zugabenvon Natriumhydroxid reagieren gelassen wird, sodaß sich Z-L-Asparaginsäure-Dinatriumsalznbildet. A process for producing ZL-aspartic acid disodium salt from fumaric acid, in which a) fumaric acid in an inert diluent in the presence of aspartase or aspartase-producing microorganisms reacted with an excess of ammonia to form ammonium-L-aspartate, then b) the solution containing ammonium L-aspartate is mixed with sodium hydroxide, the mono- or disodium L-aspartate or a mixture thereof being formed, depending on the amount of sodium hydroxide used, and c) the splitting off and excess ammonia at normal pressure and a reflux temperature of 90 - 120 ° C with residence times of 0.2 to 2 minutes continuously removed by distillation and returned to a fumaric acid suspension, which is used as a starting solution for further enzymatic reactions, whereupon d) the remaining sodium L-aspartate-containing solution is reacted with benzyloxycarbonyl chloride at a pH between 9 and 14 with the simultaneous addition of sodium hydroxide, so that ZL-aspartic acid disodium salt forms. |
