abstract |
The invention relates to a method to increase proteinnexpression in baculo vector virus expression systems. Theninvention provides a method to produce a recombinant proteinnin insect-cell culture which comprises selecting anrecombinant baculovirus expressing said protein, growingninsect cells in growth medium in a culture vessel with ansufficient volume to contain at least 2 liters and infectingnthe cells with an inoculum of at least one baculovirus at ancell density of 1 x 10 5 to 5 x 10 6 cells/ml with an m.o.i ofn<0.01. n The invention furthermore provides a method to producenrecombinant pestivirus E2 or E rns protein or fragmentsnthereof in insect cell culture characterised by a finalnconcentration of said protein (fragments) in the growth mediumnat harvest of at least 100 µg/ml. n The invention furthermore provides a method to producenrecombinant follicle stimulating hormone, α-units and/ornβ-units and complexes and fragments thereof, at anconcentration in the growth medium at harvest of at leastn15 µg/ml. |
