abstract | The present invention provides a beta -galactoside- alpha 2,6-sialyltransferase produced by genetic engineering. The enzyme of the invention may lack at least a portion of the membrane binding region of the native beta -galactoside- alpha 2,6-sialyltransferase, which is convenient for the production and isolation of the enzyme since the enzyme may be easily released to the outside of the cells when produced in host cells. The process for producing the enzyme of the invention comprises: culturing host cells transformed with an expression vector containing the DNA encoding a protein comprising the sequence of amino acid residues 16-498 of SEQ ID NO: 1 or a protein comprising the sequence of amino acid residues 16-498 of SEQ ID NO: 1 wherein one or more amino acid residues have been deleted, substituted or added, while maintaining the activity of the beta -galactoside- alpha 2,6-sialyltransferase, under conditions which promotes the expression of beta -galactoside- alpha 2,6-sialyltransferase; and collecting the beta -galactoside- alpha 2,6-sialyltransferase protein from the culture. |
