abstract |
The present invention provides a method ofnpreparing stably and at a high yield a purinennucleoside compound by utilizing an exchange reactionnof a nucleic acid base which is carried out in thenpresence of an enzyme, and also provides anmicroorganism capable of producing uracil thyminendehydrogenase or dihydrouracil dehydrogenase. Innpreparing the purine nucleoside compound, a pyrimidinennucleoside compound and a purine base are subjected tona base exchange reaction in an aqueous solutionncontaining phosphate ions in the presence of pyrimidinennucleoside phosphorylase and purine nucleosidenphosphorylase. The pyrimidine base formed by the basenexchange reaction is converted by a microorganism or annenzyme derived from the microorganism into a compoundnincapable of acting as substrates of pyrimidinennucleoside phosphorylase and purine nucleosidenphosphorylase so as to obtain a desired product ofnpurine nucleoside compound. |
