abstract | A chromatographic strip positive readout binding assay device and method suitable for quick, sensitive and reliable field testing for small molecules such as environmental contaminants, drugs of abuse, therapeutic drugs and hormones. The detectable label is not attached to either the analyte or to the analyte receptor. Low affinity binding pairs can be used in the positive readout binding assay. The strip comprises a sample contact zone, a trapping zone and a detection zone arranged along the strip in the direction of sample flow. The trapping zone comprises a binding partner in sufficient amount to bind enough of a first unlabelled binding partner so that substantially all of a labelled anti-first binding partner bound to the first binding partner is captured in the trapping zone. |
