abstract |
A method and apparatus for measuring enzyme activitynby supplying a substrate solution and an enzyme solutionninto a reaction vessel combined with a total reflectionnabsorption prism. A measurement solution (40) in which thenenzyme solution and the substrate solution are combinedntogether is kept at a constant temperature within thenreaction vessel (26) by a temperature control unit (23),nand is stirred by a stirrer (21). Infrared light (36)nemitted from an infrared light source (3) is made incidentnon the interface between a total reflection absorption prismn(28) disposed in contact with the measurement solution (40)nand the measurement solution (40) from the side of the totalnreflection absorption prism (28) and is totally reflectednby the interface. The spectrum of transmitted infrarednlight (37) thus totally reflected and emitted is detectednby an infrared light detector (5). According to thusndetected spectrum, a change in the infrared absorptionnspectrum or absorbance is determined, whereby the enzymenreaction in the measurement solution (40) is measured. |
