abstract | The present invention provides substances which have the property of activating the sequence specific DNA binding activity of latent p53, said substances being based on fragments of the C-terminal regulatory domain of p53 protein, or fragments of murine p53 including the epitope bound by antibody Pab241 (or the corresponding region of human p53). The substances are shown to activate latent p53 and to act synergistically with known p53 activators such as DnaK, monoclonal antibody Pab421 and kinases. The invention includes the use of these substances in methods of medical treatment, and in screening for mimetics and binding partners. A motif of p53 which interacts with the heat shock protein DnaK is also disclosed. |
