abstract | The present invention provides a method of nucleic acidnsynthesis capable of directly amplifying a gene of interestnin a salivary sample without purifying DNAs from the sample. n According to the present invention, in a method ofnnucleic acid synthesis by mixing a salivary sample itselfnand a gene amplification reaction solution, and thennsubjecting them to an amplification reaction, the salivarynsample is heat-treated or treated with a saccharide-degradingnenzyme before the reaction. For example, ansalivary sample was heat-treated at 0-40 °C for 1 hour,nsubsequently added directly into a PCR reaction solution,nand then subjected to a PCR. The result demonstrates that,nas shown in Fig.3, the amount of the PCR product increasednaccording as the heat-treatment temperature was elevated,nand as a consequence, the PCR product sufficient to bendetected in the electrophoresis was produced in each of allncases using different amounts of saliva, by heat-treatingnthe sample at 30°C or above. |
