abstract Disclosed is a method for determining the concentrationnof an analyte in a liquid test sample. The methodninvolves the use of colloidal sized metal particles,nwhich exhibit a spectral response in the agglomeratednstate which is detectably different than in their unagglomeratednstate, which bear specific binding partnersnfor the analyte in question on their surface. The specificnbinding partner treated colloidal particles arencombined in a buffered, aqueous medium with a destabilizernmaterial capable of destabilizing the specificnbinding partner-colloidal particle conjugates therebynpermitting the particles to agglomerate. Also includednin the liquid medium is a polymer bearing multiple analytenor analyte analog molecules along its surface whichnserves to protect the specific binding partner coatednmetal particles from dissociation by the destabilizingnagent. Analyte added to the medium, typically in thenform of a fluid test sample, competes for the specificnbinding partner and thereby decreases the amount of protectionnafforded by the analyte or analyte analog bearingnpolymer which permits unprotected colloidal particles tonagglomerate and provide a detectable spectral response.nThe magnitude of the change in spectral response is directlynproportional to the concentration of analyte innthe fluid test sample.
