abstract | The invention describes a process serving to modify the specificity and / or the efficiency of an enzyme, while maintaining its catalytic activity and characterized in that it includes: the selection of an enzyme, the tertiary structure of which is substantially known or inferred; the identification of at least one region relating to specificity and / or effectiveness; identifying or constructing unique restriction sites limiting the region identified to the corresponding DNA coding; generation of a DNA sequence corresponding to at least part of the identified region, except that the nucleotides of at least one codon are randomized, or selection as a substitute for at least one part of said identified region of a similar alternative region which can itself be similarly randomized; use of the generated or substituted DNA sequence to replace the original sequence; expression of DNA comprising the generated or substituted DNA sequence; finally, the selection of a desired modification, so as to be able to isolate the corresponding DNA coding. |
