abstract The invention relates to methods for increasing the catalytic activity of ribozymes without reducing specificity, the methods comprising contacting an RNA molecule with a ribozyme and a facilitation oligonucleotide. The present invention also relates to compositions comprising a ribozyme and an effective amount of a facilitating oligonucleotide. It also relates to methods for reducing the concentration of Mg2 + and Mn2 + required by a ribozyme to effect catalytic cleavage of a target RNA.
