abstract | The invention relates to simple lactose or lactosamine-like compounds serving as ligands for endothelial leukocyte adhesion molecule-1 (LAM-1) and related receptors. These compounds have the formula (I) in which each R1 independently represents H or lower alkyl (1-4C); R2 represents H, lower alkyl (1-4C), arylalkyl or one or more additional saccharide residues; R3 and R4 each independently represents H, (1-6C) alkyl, aryl or R3 and R4, together, form a five or six membered ring optionally containing a heteroatom selected from the group consisting of O, S and NR1; wherein said five or six element ring can also be substituted by one or more substituents chosen from the group consisting of (CHOR1) mH in which m represents 1 to 4, OR1, OOCR1, NR12, NHCOR1 and SR1; Y represents H, OR1, OOCR1, NR12, NCOR1 or SR1; and X represents -CHR5 (CHOR1) 2CHR6OR1 where R5 and R6 each independently represent H, lower (1-4C) alkyl optionally substituted by one or more F, or results in a five or six element ring optionally containing a heteroatom selected from the group consisting of O, S and NR1; said ring has five or six elements optionally substituted by a substituent chosen from the group consisting of R1, CH2OR1, OR1, OOCR1, NR12, NHCOR1, SR1 and F; or X represents benzoyl or naphthoyl comprising one to three hydroxyl substituents, provided that if X represents a hexose type substituent, R3 and R4 cannot together produce a hexose type substituent. These compounds are useful in the treatment of conditions characterized by excessive inflammation. |
