abstract |
The invention relates to a method for restoring the immunoreactivity of a tissue, in particular a decalcified tissue, fixed by an aldehyde fixing agent and incorporated into an inclusion liquid generally comprising celloidin. The method involves contacting the tissue with a solution of an aldehyde releasing reagent comprising a solvent and an aldehyde releasing reagent, removing the aldehyde released by the aldehyde releasing reagent from contact with the tissue, reacting the aldehyde in an almost irreversible manner to form a non-aldehyde derivative, and removing the excess base from the tissue. A preferred solution of decalcified tissue incorporated into celloidin comprises methanolic caustic soda at about one third of the saturation level. |
