Processes for the production in large quantities of polypeptides containing Fv antibodies, especially fragments of Fab 'and F (ab') 2 antibodies. Expression of heavy and light chain Fv in the microbial secretion system is followed by extraction of Fv from the periplasm under conditions to maintain a cysteine residue as free thiol. The free thiol is reacted with the free thiol of an antibody fragment having the same or different specificity, or with agents such as diagnostic markers or therapeutic moieties. The products exhibit advantageous characteristics of homogeneity and purity which cannot be obtained by known processes for the preparation of these derivatives. |
