abstract |
The present invention defines a useful method for sorting databases of synthetic or biological compounds according to their ability to bind specific DNA test sequences. The method is also useful for determining the sequence specificity and relative binding affinity of DNA binding molecules for any particular DNA sequence. The method involves competition in which the binding of a test molecule with a DNA test sequence modifies the binding characteristics of a DNA binding protein with its binding sequence. When such a test molecule binds to the test sequence, the balance of DNA-protein complexes is upset, causing changes in the ratio of unbound DNA to DNA-protein complexes. The test is versatile in that any test sequence can be tested by placing the test sequence next to a defined protein that binds the DNA sorting sequence. |
