abstract A process for producing a peptide or protein, which comprises producing a fused protein, which is composed of a carrier and a desired peptide or protein linked to each other via an enzymatically cleavable dipeptide such as Lys-Arg, by using a host microorganism, and treating the produced protein with at least a protease which can specifically recognize the dipeptide and specifically hydrolyze the peptide bond thereof. The fused protein includes those of a tandem type wherein the dipeptide units and the desired peptide or protein units are bonded to each other repeatedly. Depending on the breakage site of the dipeptide, the protease to be used may be an aminopeptidase which specifically liberates a basic amino acid from the N-terminus of the dipeptide and/or a carboxypeptidase which specifically liberates a basic amino acid from the C-terminus thereof.
